How to Write a Peptide Specification That Gets a Quote

The difference between a quote in the first reply and a week of questions is a one-page specification with seven fields filled in. This guide gives the template, shows each field written well and written badly, covers the private-label variant for catalogue compounds, and ends with a block you can paste straight into a Pepti-RFQ.

01

What a specification is for

A specification is the document the lot is made against and the document the certificate is read against. It is also the input to the quote: the factory prices the specification, not the conversation around it. On this desk the sales desk rewrites your request into a one-page specification sheet in the factory's format and returns it for confirmation, and the lead time, typically 15 days for a custom sequence, starts when that sheet is confirmed and payment is received. Everything you can do to make the first draft complete shortens the time before the clock starts.

A good specification is short. It does not explain the project, justify the sequence or describe the end use. It states seven things precisely enough that two people reading it would build the same lot. The template below is the whole of it.

02

The seven fields

Six of the seven feed the quote directly, as the pricing-logic guide explains. The seventh, destination, lets the factory state the shipping method and cold-chain packing in the same quote instead of a follow-up.

03

Field 1: sequence

Write the sequence from the N-terminus to the C-terminus, in one-letter or three-letter code, without spaces or with a space between every residue, but not a mixture. State the N-terminus and the C-terminus explicitly: free amine or acetylated; free acid or amide. State every modification by position, or write "no modifications". If a residue is non-standard, give its full name, not an abbreviation the factory may read differently.

If the sequence has a name in your project, give the name too, because it will appear on the label and the certificate, but the sequence governs. If two sequences are wanted, they are two specifications and two lots.

Calculate the theoretical molecular weight from the sequence and write it next to it. It costs you a minute and it is the number you will compare with the MS result on the certificate when the lot arrives.

Written well

Sequence (N→C): [one-letter sequence]
N-terminus: free amine
C-terminus: amide
Modifications: none
Theoretical MW: [value you calculated]
Working name: [name]

Written badly

"The usual sequence, amidated, you know the one."

No sequence, a modification without a position, and an assumption that the factory can read your notebook.

04

Field 2: purity target

State the HPLC purity the lot must meet, as a single target. The factory purifies to that target, measures the lot by analytical HPLC and reports the result on the certificate. A higher target means more purification passes, lower yield and a higher quote; the pricing-logic guide explains the curve. For orientation, published HPLC purity across Pepti's public COA library of catalogue lots is 98.4%–99.8% as of this page's review date.

If the project has a reason for a target, say the reason in one line; the factory may suggest a routine target that serves it at lower cost. If you have no requirement, write "routine target" and the sheet will come back with the factory's standard for confirmation.

Written well

Purity target (HPLC): routine target; confirm standard
or
Purity target (HPLC): [your figure], required because [one line]

Written badly

"Highest possible purity."

Not a target. The quote cannot be written and the lot cannot be judged against it.

05

Field 3: fill per vial

State the nominal quantity of peptide per vial. This is the number printed on the label and the certificate. Choose it from the end use, not from habit: the fill run and the label both depend on it, and changing it after the quote reopens both. If the project needs a fill that the factory does not run routinely, say so; it is confirmed at feasibility.

Note that fill is a nominal quantity. HPLC purity does not measure it. If measured fill matters to the project, add it under documents so it is tested and reported.

Written well

Fill per vial: [quantity] nominal
Measured fill required on certificate: yes / no

Written badly

"Standard vials."

There is no standard fill for a custom sequence.

06

Field 4: quantity in kits

State the quantity in kits. One kit is 10 vials. The custom minimum is 100 kits, which is 1,000 vials; the private-label minimum for a catalogue compound is 50 kits, 500 vials; a sample is 1 kit. Write the number of kits, not the number of vials or the total mass; the factory converts, but the kit count is what the quote, the packing list and the carton count use.

If you plan a repeat, say so and give the expected quantity per period. The quote can then show the lot price for the whole programme as well as for the first lot, and the MOQ guide explains why that is usually cheaper per vial.

Written well

Quantity: 100 kits (1,000 vials)
Repeat expected: yes, [kits] per [period]

Written badly

"A few hundred vials, maybe more later."

Below the minimum and unquantified.

07

Field 5: labelling

Say whether the vial carries a neutral label or your artwork. A neutral label shows the sequence or product name, the fill, the lot code and the research-use statement. Your artwork keeps those elements and adds your brand; the factory supplies the template dimensions and marks the lot-code position on the proof. If you want a kit carton or an insert, say so here; both add print and assembly and can move a catalogue order to a later lead-time tier.

Attach print-ready files at the RFQ if you have them. If you do not, say "artwork to follow" and expect the proof round to start when it arrives, not before.

Written well

Label: neutral
or
Label: own artwork attached (print-ready); carton: no; insert: no

Written badly

"Our logo on it, we'll send something."

No files, no scope, and the proof round cannot start.

08

Field 6: documents

Every lot ships with the batch Certificate of Analysis carrying the HPLC purity result and the MS identity result, a lot-coded label and a packing list. List anything you need beyond that. Be specific about the measurement, not the document name: "measured fill per vial reported" is quotable; "full documentation" is not. If you need nothing beyond the baseline, write that, so the sheet is complete.

Written well

Documents: baseline (batch COA with HPLC and MS, packing list)
Additional: none
or
Additional: measured fill per vial reported on the COA

Written badly

"All the certificates."

Unquotable, and it invites a document you will not read.

09

Field 7: destination

State the destination country and the type of consignee (research institution, company). The factory ships in insulated cartons with ice packs by courier and needs the destination to state the method and any import references required. Import permits, where a destination requires them, are the buyer's to obtain; say if you hold one.

Written well

Destination: [country], consignee: [institution/company]
Import references held: [yes/no, type]

Written badly

"Europe."

10

The private-label variant

For a catalogue compound the specification is shorter, because the sequence and the standard purity are the factory's. Replace field one with the catalogue product name exactly as it appears in the public COA library at peptioem.com/coa, choose the fill from those the product is made at, set the quantity at 50 kits or more (or 1 kit for a sample), and attach the artwork. The sales desk returns the sheet with the standard purity filled in for confirmation. Standard lead-time tiers are 3–7, 10–14 or 14–18 days from confirmation and payment, and the quote names the tier.

Order the sample first with a neutral label; it arrives with its lot certificate, and the production order can then repeat the configuration exactly, which keeps it in the fastest tier the quantity allows.

Mistakes that send it back

  • A sequence without termini. Free acid and amide are different molecules with different masses. The factory will ask.
  • A modification without a position, or an abbreviation with more than one meaning.
  • "Highest purity" instead of a target, or a target chosen from someone else's certificate rather than the project.
  • Quantity in milligrams or "vials" without a fill, or a quantity below the minimum written as if it were negotiable.
  • Artwork described but not attached. The proof round starts when files arrive.
  • "Full documentation" instead of a named measurement.
  • A destination region rather than a country.
  • Several sequences in one message without saying whether they are alternatives or separate orders.

None of these is fatal. Each costs a round of questions, and the lead time does not start until the round is closed.

The paste block

Copy the block, replace the brackets, and send it as the body of a Pepti-RFQ on WhatsApp or by email. If a field does not apply, write "none" or "confirm standard" rather than deleting the line; an empty line is a question, a filled line is a specification.

What happens after you send it

The sales desk writes the request into the factory's specification sheet and returns it with a written quote. For a custom sequence the quote carries the expected lead time, typically 15 days from confirmation and payment, and any feasibility notes; for a catalogue compound it names the lead-time tier. You confirm the sheet, or send all corrections at once, and pay. From that moment the process page describes every step with its owner, and the receiving checklist describes what to do when the carton arrives.

FAQ

Questions buyers ask

Do I need to send the sequence before I get any answer?

For a custom quote, yes. The sequence determines synthesis difficulty, purification and the identity check, so no quote can be written without it. For a catalogue private label, the product name is enough.

Can the sales desk fill in the fields I am unsure about?

Yes, if you say which ones. Write "confirm standard" or "propose" on the line and the sheet comes back with the factory's usual value marked for your confirmation. Leaving the line blank produces a question instead.

How should I state a modification?

By position and full name, for example the terminus or residue number and the modification applied. Also state the termini explicitly, because free and amidated C-termini are different molecules with different masses on the MS result.

What is the minimum quantity I can put on the sheet?

100 kits (1,000 vials) for a custom sequence, 50 kits (500 vials) for a catalogue private label, and 1 kit for a sample of a catalogue compound.

Is the sheet a contract?

The confirmed sheet is the document the lot is made against and the certificate is read against, and the written quote references it. Changes after confirmation are handled as amendments, which can restart the affected steps and change the quote.

Request a quote

Send a Pepti-RFQ for your specification

Open WhatsApp with a pre-filled Pepti-RFQ message, or email the same details. A quote needs the compound or sequence, fill per vial, vial count, labelling and the document set you require.

  1. State the sequence or catalogue compound and the fill per vial.
  2. State the quantity in kits (1 kit = 10 vials) and the destination country.
  3. List the documents you need with the shipment.
